Despite being a cosmetic concern, discoloration doesn’t start on the surface. It begins deeper, where oxidative stress disrupts pigment regulation long before a dark spot appears.

When your skin is exposed to sunlight, pollution, or inflammation, it produces reactive oxygen species (ROS). These unstable molecules disrupt the way your skin regulates pigment and slowly break down collagen, eventually leading to visible changes that can make you appear older than your age.

It’s a slow progression, which is partly why treating it can be so frustrating. Most brightening products focus on correcting what’s already visible. However, new research suggests that earlier intervention—before ROS can cause damage—may hold promise for a brighter future for those living with skin discoloration.

The Internal Imbalance That Leads to Lasting Discoloration

Before we introduce you to ProteXidine™, let’s review how discoloration happens in the first place.

When your body produces ROS in response to oxidative stress, it interferes with multiple systems at once. One of the first to be affected is melanogenesis, the process that controls how your skin produces and distributes pigment.

Under normal conditions, melanin is created in response to UV exposure to help protect the skin. That’s why, for example, people who live closer to the equator tend to have darker skin and eyes, while those in northern climates—where sunlight is limited—are more likely to have fair skin, light eyes, and blonde hair.

However, when ROS are present in excess, they ramp up the activity of tyrosinase, the enzyme that acts as the catalyst in melanin production. This overstimulation disrupts the skin’s internal feedback loops, leading to excess melanin in the wrong places. Over time, that imbalance creates the uneven tone we recognize as hyperpigmentation.

ROS also weakens the structural proteins that help the skin recover from that disruption. Collagen fibers begin to break down faster than the body can rebuild them, and elastin fails to form the neat matrix pattern it typically does.

By the time pigment actually reaches the surface of your skin, multiple systems that keep your skin balanced and self-correcting have been interrupted. That’s why, as you age, hyperpigmentation tends to last longer, and typical brightening products may not be as effective.

Interrupting the Cascade with ProteXidine™

ProteXidine™ is a patented biomimetic decoy molecule developed through a decade-long research partnership at the University of Minnesota’s Center for Drug Design. It was created with a singular goal: to prevent the formation of ROS before they can trigger the chain reaction that leads to discoloration. ProteXidine™ caught the interest of TOPIX Pharmaceuticals, Inc., which the U of M then entered into a licensing agreement with.

Most antioxidant ingredients work by neutralizing ROS after they’ve already formed, at which point the oxidative cascade is already in motion. ProteXidine™ is different because it helps prevent their formation altogether.

Its structure allows it to act as a molecular decoy, mimicking the proteins and lipids that ROS typically target. When those unstable molecules bind to ProteXidine™ instead of skin cells, the chain reaction is interrupted before it can lead to excess melanin, inflammation, or collagen breakdown.

“Sunscreens are very good at shutting down erythema (reddening of the skin),” explains Steve Hernandez, senior vice president of R&D at TOPIX Pharmaceuticals. “In other words, when you put sunscreen on, and you’re not going to get a sunburn for much longer than it would take without it. Therefore, you feel that you can safely extend your time in the sun. But, while you’re not burning, you are still damaging your skin and generating these markers of high concern in your DNA molecules. These are called Cyclobutane Pyrimidine Dimers [CPDs] also known as thymine dimers in the DNA molecule.”¹

In lab studies, skin treated with ProteXidine™ showed a reduction in the formation of CPDs—tiny lesions that form in DNA after UV exposure and contribute to photoaging. The ingredient also appeared to help skin clear out damaged cells more efficiently, speeding up the turnover of pigment-prone areas so that new, healthy cells can take their place.

AST Brightening & Firming Complex: Medical-Grade Skincare to Fight Discoloration

Every active ingredient, including ProteXidine™, was chosen for its ability to support the skin through each stage of the pigmentation cycle, beginning with prevention and continuing through long-term recovery.

  • Hexylresorcinol helps reduce the appearance of visible discoloration while calming the irritation that can lead to issues like post-inflammatory hyperpigmentation.
  • Arbutin, a well-studied plant-derived compound, slows melanin production in areas where pigment has already reached the surface but without interrupting your skin barrier.
  • Hexanoyl dipeptide-3 supports healthy cell turnover without the need for harsh exfoliants by replicating the skin’s natural exfoliation signals, encouraging pigment-heavy cells to shed naturally. This more gradual exfoliation helps prevent dead skin cell buildup—which contributes to dullness—and lowers the risk of rebound discoloration, particularly where the skin barrier has been compromised by inflammation or exfoliation

“UV is a free radical chain reaction initiator,” Hernandez explains. “Free radicals have unpaired electrons, meaning that they’re highly unstable, and want to return to their ground state as soon as possible. And to do that, they will deteriorate and destroy the things around it. These chain reactions are explosive, because each free radical triggers others to form. That’s why we love antioxidants, they have a rich pool of electrons for free radicals to take from, thereby stopping that shotgun blast chain reaction. The antioxidant takes the bullet for you.”¹

“Your first line of defense from the sun is SPF,” Hernandez says. “You can think of antioxidants as a kind of second line of defense, but they aren’t going to trap all of those free radicals. Our clinical data suggests that ProteXidine™ allows your body to make more repair enzymes faster, thereby providing another line of defense against the sun.”¹
